Sir I isolated Polyhydroxybutyrate(PHB) producing bacteria i.e Bacillus sp. and started extraction of of PHB. Sir I want to do characterizaton of the extracted PHB by using of FTIR,HPLC,GC-MS,LC-MS. So, I want standard for characterization. From Sigma Aldrich which one you will prefer for standard for comparsion with my extacted PHB.
Dear Sir. Concerning your issue about the use of Poly[(R)-3-hydroxybutyric acid] natural origin of Sigma Aldrich as a standard for characterization PHB extracted from Baciilus sp . Of course you can use it as standard. I think the following below link may help you in your analysis:
You may try one of the following protocols for PHB determination?
"Biomass composition and methods for the determination of metabolic reserve polymers in phototrophic sulfur bacteria"
"Gene regulatory and metabolic adaptation processes of Dinoroseobacter shibaeDFL12T during oxygen depletion"
"Metabolic network analysis of the marine bacterium Dinoroseobacter shibae" (page 37)
You can obtain PHB from Sigma-Aldrich (http://www.sigmaaldrich.com/catalog/product/aldrich/363502) to generate HPLC standards.Good LuckMichaelhttps://link.springer.com/article/10.1007/BF00877431http://www.jbc.org/content/289/19/13219.full.pdfhttp://scidok.sulb.uni-saarland.de/volltexte/2016/6473/pdf/Dissertation_Annekathrin_Bartsch_Endfassung.pdf
