Is it possible to polarise CD4 T cells to obtain homogenous CD4 Treg population ?

Polarisation of CD4 T cells from splenocytes of OT II mice for 6 days does not result in homogenous CD4 Treg populations.

The polarisation was done by addition of IL-2 and TGFB on day 2 and day 4.

Does anyone has experience with this ?
