Hi. I'm growing Arabidopsis thaliana on ágar medium (0.8%), aiming to extract some enzymes of the seedlings 120 hours after germination (just 5 days). Do I need to prepare the MS medium? Or can I just grow the seeds in the agar medium without the MS medium?
Why not give it a go and see how well they grow? It will only take 5 days, and if the seeds aren't able to support themselves for 5 days' growth, you might need to add a little bit of nutrient in the form of dilute MS medium.
Still, I'd be fairly surprised if the seeds were so depleted of nutrients even after multiple generations of culture in a laboratory setting that they couldn't make it to 5 days old.
I'm agree with Matthew, 5 days in pure agar medium will force seeds to release enzymes and seedlings intensively use nutrients accumulated in the seeds, addition of MS may change enzyme profile, it is similar to roots behavior in soil or culture they take up nutrients from growth media after almost full use of the nutrient, starch, fat, proteins from the seeds
