Is it possible to determine mechanical properties of filler in powder state?

I would like to know how to determine mechanical property of filler in powder form

If you only have powder to test then I'm not sure what you could do. Exactly what properties do you want to measure?

If you can put the filler into a matrix then you can test a part made of filler+matrix and another part of matrix alone. Knowing the volume fraction of the first test piece and the stiffness of both then you can estimate modulus of elasticity of the filler.
