What are the other parameters which should be optimized?
I have seen that people directly use solution of particular concentration and do the electro-chemical experiments.
I have observed this in Very high impact journal like in Biosensors and Bioelectronics also.
Is it right? if yes then why?
It is very important, because depending of your film you can change the surface charge, or even find a pH where you enzyme do respond better, usually for glucose oxidase pH around 7 is used, but for other enzymes and films this kind of experimental routine is really needed.
It is advised to carryout sensing experiments in physiological pH around 7. However you should observe the current response obtained from other pH values too. Increased current response will be seen for any particular pH which depends on the modified medium that you are employing. In addition for sensing other parameters such as potential applied, response time etc should also be taken into account.

For Enzyme based sensors, the pH of the experimental could be optimized to define the pH at which improved performances occur. Obviously, physiological pH is preferred for practical applications, such as analysis of glucose in physiological fluids, pharmaceutical preparations etc.
However, even with non-enzymatic based sensors also, optimization of pH is very much preferred. Specifically, if the electrochemical sensing mechanism of glucose involves an uptake or release of protons, then the localized pH at specific points and portions of the sensor chip could be very different from the bulk pH. Therefore, optimization of pH specifically using buffer solutions could be preferred irrespective of enzyamtic or non-enzymatic processes.
