Im working with soy lecithin and have problems with film formation .so lipid seem to be separated at the end of rehydration process so does phase transition temperature is affecting? And if yes than what precautions should I take ?

If I understand your question correctly then indeed the transition temperature is important in this matter. The thing is you need to be either below or above the transition temperature, for example a sonication procedure (a way to create liposomes) is often performed on ice to ensure you remain below the transition temperature. The thing is that when the mixture is at the transition temperature (or is going through the transition temperate) all sorts of membrane disruption (causing for example leakage) processes take place.

After organic solvent evaporation, you should increase the temperature above the transition temperature before adding the aqueous solution. And the added aqueous solution should be at the same temperature and added slowly on the flask wall not on the film directly. You can know the transition temperature of your lipid composition by DSC or by raising the temperature until you see the premelting of the lipid.
