I have prepared solid dispersion nanoparticles by solvent evaporation method but the practical yield is very low,so i am facing do calculation error?

I need to find out the in vitro dose,but due to loss, i am not able to calculate that, as per theoretical values,the release is coming in negative values.can anyone facing same problem or have any idea about
