For standard curve, the cholesterol was dissolved in chloroform and then reacted with LB reagent and was fruitful. But when cholesterol was dissolved in acetone and reacted with LB reagent, an exothermic reaction was observed with pale greenish appearance. Further more cholesterol was dissolved in DMSO and reacted with LB reagent, this time no apparent changes in reaction mixture were observed. Another problem I am facing is chlorophyll in chloroform extracts. I think the chlorophyll content will interfere with final results of the samples, what do you think?
100mg powdered plant samples were extracted with 600 micro-litter of acetone and than 10mg of the final extract was dessolved in 100micro litter of DMSO for phytochemical screening, so i want detect phytosterol in these extracts.
I am also agree with Olga Lock, use chloroform solvent for sample preparation
