Dear researchers, I will work on plant extract. I will seperate the plant extract with sephadex. Could I get information about this method? How will I use the sephadex? Is it necessary to dissolve in buffer? Thanks in advance..

You need to dissolve resin because different types of sephadex have different degree of swelling and molecular fractionation range

Dear Selvinaz-Hancerli,
Sephadex is supplied as a dry powder and must be swollen before use (H2O or buffer like PBS). Avoid excessive stirring during swelling as it can break the particles. Do not use magnetic stirrers.
After this use column and buffer for equalibration
