I'd like to perform single-strand conformation polymorphism (SSCP) in my thesis, however I cannot control the temperature of the vertical PAGE since we are using the conventional tanks. Is there a way that I could somehow lower and monitor its temperature?
The simplest way to lower the temperature of the gel is to run it for longer but at a lower voltage. Better still is to run it in a cold (refrigerated) room if you have one. you will get some cooling by aiming a fan at the plates to help shift some heat. I do not know about monitoring temperature but modern skin sensing thermometers or possibly some smart phones have temperature apps which may not be very accurate but are probably reproducible. All methods are approximate since the gel is thin and it loses heat into thick glass plates so you are really monitoring the plate temperature and the gel temperature will be higher
I used to find that better results were achieved in gels with 5% glycerol added
