I want to estimate ammonia from the reaction mixture of my interest. For that purpose I have to first prepare a standard curve of ammonia. I go through the literature for the purpose and I found that most of the researchers used Nessler reagent to do that. But I want to know the exact protocol for the determination of ammonia by nessler reagent. Please reply if you have the specific protocol or give me some link so that I can find out in internet.
Use ammonium-sulfate or ammonium-chloride for standard preparation.
Because Nessler reaction is very sensitive appropriate dilution must be used.
Recently we calibrate as follows for ammonium-nitrogen:
Range: 0-50 ug NH4-N
3 ml of std + 2 ml Nessler-reagent (made in-house without any base ) + 3 ml NaOH (2M)
It is linear with r2 > 0,98, max abs is about 1,2 A unit.
Yet it is not well established, recently I try to optimize it, but works good.

How to prepare standard curve, I mean what should be the concentration of ammonium sulfate? and what is the range of ammonia detection by Nesslerization method using spectroscopy.
