Hi,
i synthesized cyclic peptide including disulfide bond from linear peptide.
However, I could not find a way to check whether the synthesis was successful.
In most cases, it is confirmed by mass spectroscopy(MS/LC), but I don't have access to that method.
How can we distinguish between cyclic and linear peptides except MS/LC?

Have you tried paper chromatography, that is a powerful method for peptide determination + it can be non-destructive.
Ninhydrin or silver or iodine or UV visualization.
If you don't have paper you can also try reverse phase TLC plates (paper is usually better).
First off, I would suggest a good column separation or HPLC separation and then use NMR for the structure characterization since within cyclic structures closer hydrogen atoms will shift greater than an aliphatic-acyclic structure. Hope this helps.

Although I had access to mass spectroscopy to make the linear and cyclic peptides, but it is time consuming to get data from mass spectroscopy. As a result, I have sometimes used the Kaiser test which was utilized to ensure that each coupling reaction was completed. Let me know if you need more details about the Kaiser test. It is a very popular test for the peptide synthesis and you can find the details of the test in many scientific articles. I hope it helps.
