To put it simply, if there are no peaks, there is nothing that any kind of peak potential would refer to. There is no (redox) electron transfer, hence, there is no rate to determine - assigning any peak potential / rate constant values would, for a CV lacking any peaks be meaningless.

While broadening the range of applied potentials (and possibly choosing an adequate potential scan rate) will likely allow you to find peaks, I would reocmmend that you first do the following steps:

1. Identify the redox reaction that you wish to investigate,

2. Based on literature reports, attempt to predict, at what potentials and in what conditions should you see a peak related to that reaction.

3. Verify assumptions experimentally.

4. (if necessary) Alter experimental conditions and repeat step 3

In my opinion there is a high barrier to electron transfer, leads to recession of the electron transfer reactions. In this case the system become electrochemical irreversible with no observed oxidation and reduction peaks. So, more negative/ positive potentials are required to observe the reduction/ oxidation reactions.

Now is the biofilm on the electrode play roll in this case?

The problem I am facing is in the determination of Ep value from a voltammogram with no peaks. I am interested in calculating the electron transfer rate from CV data.

Sometimes we don't get required peaks due to less residence time at high scan rates. At high scan rates, the electrolyte ions don't get much time to interact with the redox species available inside the pores (micropores, mesopores, nanopores, or any; depending upon the texture). You can try the experiments at even low scan rates (though you've already performed at low scan rates). Potential stability window is another important criteria in cyclic voltammetry, which has to be taken care of while performing CV.

Without any peak in CV, you can't propose redox potentials of your prepared material. You can follow different literature related to your material to have an idea over the redox potentials of the corresponding species.
