I'm doing a research "perception towards programming based on gender." I've written the null hypothesis
H0 : Both male and female students equally likely believe that programming course is important for future career
I'm not sure about the alternative hypothesis. It is possible to conduct a two tailed test but I feel by conducting a one tailed test I can derive more information. so for alternative hypothesis I can write either
H1 : male students believe more than female students that programming course is important for future career
or
H1 : female students believe more than male students that programming course is important for future career
but do they make any difference in calculating test statistic in spss?
H0: there will be no differences between genders in case for an attitude for programming.
H1: there will be significant difference between male and female students in such attitude.
your next hypothesis caould be more precise as you described as "H1"
So you can check if the gender have an effect (two tailed), as well as if males will have an higher scores than females (one- tailed). Usually Mann-Whitney is performed as a two-sided test. So basically everything depends on your hypotheses.

Thank you for the clarification. I carried out the hypothesis as you said. H0 is rejected. now can I carry out Mann-Whitney again as a one tail test to check male have a higher score than females?
