I would like to incorporate the active substance in Lipid Nanocapsules (LNC) formulation. Unfortunately, I don't know how much the amount of the active substance would be needed to add to the LNCs formulation. I have the IC50 values of free substance and blank LNC (without any substance) in the breast cancer cell line. Could anyone give suggestions on how to calculate the amount of active substance that would be added to LNC formulation based on the data that I have?
I really appreciate any idea that you suggest.

Your loaded Lipid Nanocapsules must be centrifuged at 10,000 rpm at room temperature for 1 h. Collect the drug contained in the supernatant and quantified using HPLC .
Calculate the Encapsulation efficiency (EE) and loading efficiency (LE) of drug using following formula;
EE= [(Wt-W)/Wt] x100 %, where Wt-mass of your drug after concentration, W-mass of drug before concentration.
Mass of loaded drug (Wd)= Wt-W, Mass of Loaded drug and lipid nanoparticle =W0
LE=(Wd/W0)x100 %....
