You can combine the mass of cathode and anode to determine the discharge capacity.

I would recommend to measure it with a regular charge/discharge test.

Theorethical calculations lack some effects.

 Hi, first: Full charging the cell using CC-CV method at 23 °C. Second, dirscharging the cell with 1C at 23 °C until the lower voltage Limit is hit.  The obtained Ampere-hour or Ampere-second value is equal to your cell full capacity. In this regard, following Parameters mut be known: Upper and lower voltage Limits; Nominal capacity of the cell. Best regards

yes you can take mass of cathode not anode. lithium ion full cell capacity is dependent upon specific capacity of cathode mAhg-1and it's mass.
