I am trying to measure cortisol using HPLC/MS. I cannot develop a standard series since the AUC results for my various cortisol dilutions are not proportional. The calculations are correct. I diverted back to only trying to measure 1:10 and 1:100 dilutions of my cortisol stock. STILL, the AUC results are not comparable. I.e. 1:10 (200 000μg/L) turn out with an area over 24.000.000 and 1:100 (20.000μg/L) with an area of 5.000.000.

I tried various set-ups: 35-50-85% of organic solvent (either ACN or MeOH), C18 and Phenyl-columns with and without precolumns, dissolving my stock in/diluting it with MeOH, H2O, or mixtures of both. I am out of ideas about what could solve my problem. Has anybody encountered similar difficulties or have any ideas of what the problem could be?
