I plan to grow a probiotic culture in MRS Agar in a flask and I am wondering how do I transfer the grown culture to a tube without destroying the cells, for centrifugation.
or should the culture be grown in a broth instead and after growth the same broth tube will be the one to put in a centrifuge?
I apologize firstly for my lack of knowledge in this field. I am still trying to familiarize myself with cell culture basics but I am doing my best to learn. I would really appreciate some help.
Usually for that purpose you can grow in liquid broth and spin down the culture directly after it had grown (transfer the culture to a 50ml falcon).
You need to opt for growing in a Liquid medium in the absence of Agar. If you have the strain in a plate, you can inoculate it into a liquid media. You can spin down in the same tube, or even transfer it to a falcon for centrifugation. Make sure to handle cultures inside the LAF and ensure sterility. I personally prefer growing in glass containers and then transferring them to a falcon for centrifugation.
