I am using the spectrophotometer method to determine total lycopene in tomatoes, using the formula
C(mg/kg fresh weight) = vol of solvent/weight of sample * absorbance * 3.5094
The absorbance reading at 503 nm was very high so I diluted 1ml of the extract using 5ml of the solvent before reading the absorbance.
How do I incorporate this dilution carried out into the formula for calculating the lycopene content.

You should multiply your new result with dilution factor which is the ratio between the final volume of diluted solution (6ml= 5 ml solvent + 1 ml extract) and initial solution (1ml extract).

What is the other product for a dilution of lycopene without using chloroform, since the latter is cytotoxic for use in cell culture? How to do this dilution?

Lycopene is soluble in organic solvents and other hydrophobic materials as oils. I think it is difficult to deliver lycopene to the cell culture which is water base. You can prepare emulsions w/o or o/w with lycopene solved in oil.
