i carried out ethanol extraction of my plant extract using soxhlet method. Thereafter, i did column chromatography ,using six solvents to elute..Using standard tests for alkaloid , it was confirmed to be present in methanol and water fractions but my challenge has been how to crystalize my alkaloid out of the solution. How do i get my alkaloid out of the solution?

Either you can try with ammonium hydroxide solution.
or by dialysis of aqueous solutions of their salts through a semipermeable membrane
The first step is to evaporate to dryness those fractions containing the alkaloid. If you have eluted with pure methanol and water then there will be no residue other than your compound. Now you must attempt to find a solvent in which the alkaloid is soluble. I would try ether (diethyl or propyl) or a chlorinated solvent. But not ammonium hydroxide. Once the alkaloid is dissolved, you can use standard crystallisation techniques to isolate crystals. I would start by leaving the solution to evaporate slowly.

Accidentally I found a method that alkaloid could be crystallized finely. By using ethyl acetate, ethanol, hexane, and ammonia solution 25%, 100:5:5:2.5. I left it alone, and found crystallization after several days.
