I need to transport 4%PFA-fixed whole murine brains between collaborators (Swiss-UK) so that, in an ideal world, I can cryo-section them. So far, we have tried transport in PBS, however I find the quality of the tissue sections is very poor (in certain brain regions) in comparison to a brain that has not been transported. I can only presume a change in temperatures during shipping has caused a problem, as the shipping time itself was under 48 hours. Has anyone found a simple method for transporting brains which maintains the integrity and quality of the tissue observed after cryosectioning? e.g. Will transporting in sucrose be better? Does anyone transport O.C.T embedded tissue? Does anyone dehydrate their tissue for transport, and rehydrate before cryopreserving and sectioning? Do people transport their sections? Any experience/insights would be very helpful.
