I want separate natural product (flavonoid) from plant materials. I separate components of plant methanol extract of plant by analytical HPLC but not very good. How can I scale up analytical to preparative HPLC? Is there a equation that calculate scale up that include all parameter (length, diameter, particle size, sample amount injection, flow rate) influence on spectrum?

To scale up to prep HPLC you may need a prep system (an analytcal system may not be suitable for a prep column, quite expensive, by the way, because of the different flow rates, loop....). But you could use your analytical system with a semi prep column using the same HPLC method: with a series of injections of 100 ul you might be able to obtain some of your product,, enough for an H NMR for example (you might have a worse separation with a bigger load:you cloud check whether you are still happy with your peak resolution..). If you do not have a fraction collector you could collect the fractions manually (very long and tedious job but doable).
