I have some exeperinces about plant tissue culture.
I know that type of explant is very important to set up your experment on tissue culure
as you know,
you can have callus from any part of plant
so,
in my study, according to the target of my study, i will choose the suitable explant.
but
in most casses
we use hypocotyl or cotyledons i available, it is thought to be more easy

I found this paper but I think this is not exactly answer.I sent mail for this person but she didnt answer.I tested many experiments on R.foetida with different explants and different Hormone concentration but it doesnt reply.

Have u ever tried the other type of media?
u could also find another species close to ur plant and apply exact media but with different concentration of PGR's.
I used different PGRs but it doesnt grow.I am using this media for R.canina and R.beggeriana.both of them are good and both of them are wild roses.
