I am working on surface functionalization of calcium phosphate nanoparticle using APTES with ethanol as a solvent. Later on I'm planning to attach proteins on it. I've tried to sonicated the sample before and after the condensation as pre-treatment and to remove physiabsorbed amines, and I washed it around 5 times using ethanol. But it seems to be produced microns in size. Could you kindly provide me with suggestions on this matter?
So you started with nanoparticles and ended up with microparticles? APTES must be added in excess with respect to the area to be covered of the calcium phosphate nanoparticles. The reaction must be heated up to favor APTES condensation, you can also add some water in this regard. I do not think you mean physiabsorbed, perhaps physiadsorbed?
I am not sure if APTES functionalization has a great effect changing nanosize to microsize. This may happen if you have low charges of nanoparticles, then the repulsion of every particle is low. Consequently, aggregation may happen.
You could measure the zeta potential first.
Several studies mentioned that using dry toluena for APTES is better for functionalization.
