I will use caspase-3 kit (BML-AK700 Enzolife) for rat brain hippocampus and retina tissues. But I don't know which solution I should use for tissue homogenization, PBS or another solution? I could not find any information in the kit paper.
Clearly, the answer should be PBS with at least Three wash and BSA-TBS for negatives control.
I suppose that you can also stain the slices with DAPI.... same procedure
NB General comment: It is totally scandalous that the Kits are very expensive in general and do not contain original substances. The secret keep around the kit composition is just a matter of commercial potential... Especially the Kits from ENZO.... I would like to start a discussion on how we can dissect KITS and propose to the users an alternative which is to buy the components separately to a much lower price.
