I have read a paper that stated that after 30 minutes sterilization of 96 well microtitre plate with UV from biosafety cabinet, no cfu was recovered from biofilm of E.coli. However, I sterilized for 1 hour and then add fresh media to resuscitate possibly damaged bacteria and incubated at 37 degree for 1 hour and bacteria can grow back. Is there a better way to sterilize these bacteria inside biofilm without damaging the plate and biofilm structure?
Hello, actually biosafety cabinet UV intensity is not less 40 mV/sm^2 which enough for bacteriostatic effect but not total bactericide effect. Today already certain number of bacteria resistant to UV.

I think untill now no approved method to sterilize these bacteria inside biofilm
Seems like UV radiation presents poor penetration in the matrix of biofilms, may affect only top layers of microbial cells are exposed to its deleterious effects. Here are articles for more details

Sunagar Raju I have tried sterilization of E.coli biofilm as mentioned by Tingpej et al but it is not working. May be because I use a flat bottom 96 well plate while the paper use calgry device. I have tired up to 1 hour under UV in biosafety cabinet but not working. May be I will combine using disinfectant and UV and see if that work or will prolong the exposure time to 2 hours and check the result.
