I want to use different concentrations (1*10^4; 1*10^5; 1*10^6; 1*10^7; 1*10^8, and 1*10^8) of fungal conidia for bioassay. Can any one provide me with a detailed explanation on standardizing the conidia into these concentrations.

Prepare a serial dilution of spores in broth. Determine each liquid's 'optical density' as per this article (may be adjusted if necessary). Plate 0.1 and 1.0 mL of liquid on PDA agar (Potato Dextrose Agar). Incubate 5 days at 25 Celsius (Carbon Dioxide atomosphere if anaerobes). Count and plot curve!
