Can anyone tell me about the heat sensitivity of aqueous solution of AgNo3? I have to use AgNo3 in MS media as ethylene inhibitor but a little bit confused how to use it should I autoclave it with media or add after autoclaving?
As advices by Mr. Singh , You have to add silver nitrate to the MS after autoclave and when it cools just before you pour the solution to plates. You can leave the magnetic stirrer inside the bottle for autoclave and add the solution to stir the media. As for the prep of silver nitrate solution, you can filter sterilize using millipore syringe. Hope this helps.
The issue with silver nitrate in MS salts really isn't heat sensitiviey at all. It is a question of solubility. I think you will have difficulty knowing what the actual concentration of Ag is in your solution. MS salts has both significant chloride and phosphate levels, and thus AgCl2 and Ag3(PO4)2 percipitates will form depending on pH of the medium. Therefore you will likely have a very low silver concentration in the medium, which may or may not be effective regardless of when in the media prep process silver is added to MS salts.
