It is highly dependent to the type of response of your polymer and pH which your polymer change its properties,

Its may be tuned with stomach acidity for oral drug delivery

And may be work with endosomal pH, which is usefull for gene/drug delivery

And even may be tuned with tumor microenvironment.

But for invivo application you can perform following test

1- design a similar polymer whitout pH responsive part to be used as control

2 labelling the polymers with an near ir dye to be able evaluate the efficiency of polymer

3 perform test in perscence of some component which change pH for example neutralize the acidicenvironment
