The assay can be done by MTT, LDH, Alamar Blue and others. Normally, you should perform at least 2 of these because most  of times, you have some absorbance  influence of the nanoparticle. You can also use primary cells instead of this epithelial cell lines. Maybe, the attached paper can help ypu.

Flow Cytometry is best way to find the toxicity.

What kind of nanoparticles are they - metallic, polymeric, SLN, liposomes etc? Several literatures have different techniques to determine cytotoxicity of drug loaded nanoparticles depending on their type. The above papers are good one.
