I am planning to analyze proteins by using NMR to know what are they. Most of them have about 5 kDa. Could you recommend/advice any protocol or method how to prepare proteins for the analyses?
Best is to look at the sequence and the corresponding NMR Signatures of the Amino Acids. Analysis of Proteins would require 2D or 3D NMR analysis Technique, Your Regular COSY or TOCSY Experiments can shed some light on the Conformational Stability etc. of your proteins. Therefore Start from the Basic and depending on what you are looking for increase the complexity of the NMR (COSY-TOCSY-HMBC-HMQC-NOESY-ROESY) so on and so Forth. With variations in the experimental parameters in T1,T2, T1*, Concentration of Protein, Tempeature etc you can really delve deeper into the conformational and stereochemical aspects of the Proteins.
