I have recombinant form of bacterial type 3 secretion system effector (protein), which is believed to form pores in artificial bilayer membrane (in-vitro). But the protein is unable to make pores, can anyone suggest me some optimal conditions to enable this protein for pore formation in the membrane. Does expression in E.coli might have any effect on its pore forming capability ?. Thank you
