We are working on an aquatic plants and had lot of problem in cutting them because of their water content.
sorry it was to cut the leaves to observe chloroplasts..etc under fluorescence microscope with an aquatic plant that has thick leaves
Thanks indeed a razor was used to cut the leaves but the results are not fantastic as still very thick
Dear Brigitte,
I would ask silvia.mazzuca who is an expert in Marine Plants and extracts chloroplasts from theses species.
Norbert
Dear Norbert
Thanks a lot for this information.
Best wishes

Dear Brigitte
I hope the reference below can help you.

we want to observe autofluorescence of chloroplasts and to use some specfic fluoresencent dyes as well to merge pictures
maybe someone in the group of Hendrik Schubert (University of Rostock) can help you? he has done a lot of charaphyte research.
Thanks for this advice.
Assuming the leaves are soft you can cut nice sections with a vibratome. You should be able to use either fresh or fixed tissue for this.
