I work with the Folin Ciocalteu assay and developed a microplate method. Basically in a 96 well plate, transfer 10ul of alcoholic/or aqueous plant extracts in wells. then put in 100ul of Folin Ciocalteu reagent (diluted 1:10 with distilled water) and 80ul of 1M sodium carbonate. Incubate at room temperature in the dark for 20 min. Then read absorbances on a microplate reader at a wavelength preferably 750nm or 630nm.

You need to set up a calibration curve with gallic or tannic acid. Use 10ul per well here too+100ul FCR and 80ul Na2CO3. I usually go for the following dilutions: 960 ug/ml, then 1 in 2 dilutions 480, 240, 120 and 60 ug/ml. I use distilled water as the 0 ug/ml.  From y = mx+c you can convert the extract absorbances into gallic acid equivalents in ug/ml.

Use three wells per extract/standard so as to have triplicate values.
