If I prepare my sample in PBS, do the salt in PBS will affect the TEM image quality?
Salts form crystals, which may overlay your specimen.

after fixation you have to wash with water before dehydration. without washes, it may happen to observe samples with saline precipitates

I agree with Simone Cantamessa, If you use PBS, you need good washes with water to avoid crystals.
Please inform me if any one have SKBR3 cells in Indian laboratory and kindly provide a flask or vial. It is very much urgent, your help will be appreciated.
