The buffering range of PBS and BBS seems to be quite similar to me. However, from literature it seems that many of the neuronal recording (both electrophysiology and FRET) are done with BBS. Is there a specific reason for this?

If recording is made from cultured neruons, the PBS or HEPES buffered solutions are fine. When you are about to record from acute brain slices, the bicarbonate buffer must be used because the aCSF needs to be oxygenated with 95% O2/5% CO2. Bicarbonate and CO2 are used to keep the normal pH of the solution.

Thanks Xiao-hui. Is it because the pH changes caused by 5% CO2 could only be buffered by bicarbonate and not by phosphate system because there is no conjugate base for CO2 in phosphate buffering?
