I used phenol extraction protocol to extract protein from A. marina root for PAGE. However I did not get enough protein and also the pellet color after ammonium acetate precipitation was dark brown. I need to modify the protocol for more efficient protein extraction considering high phenolic compounds in the A. marina root.Any suggestions welcomed.
Try to investigate the solubility of other previous research articles first.
Try water ,alcohol in different concentrations such as 40% 50% or 80%, also ultra sonication , or organic solvent.
If its just for checking SDS-PAGE pattern, then you can try extracting directly in SDS or SDS-PAGE sample buffer.

The problem is not about PAGE. The root protein content is much less than shoot. I want to have protein profile of root under stress. I don't want to loss protein because of extraction procedure. need to use an efficient protocol.
