I am conducting an ethnographic study where I collected data via participant and semi-structured interviews. At the beginning of the research, the ratio of male and female participants was OK. However, when I came back for my interviews (after 6 months) there was only 1 female to interview (before they were 6). My supervisor thinks that this is not a meaningful observation. How can I convince the readers that it is meaningful?

Everything is meaningful in research as long as we present our case accordingly. In your case, you need to present the sound logic of reduction of female participants or may be in later stage if you dont find any, you can present this as the limitation of the study stating the attrition of sample.

It is no clear from your question:
1) what was the total number of persons interviewed in the first round, perhaps females was a small share in the first round?
2) Anyhow, the lost of interviewees by 20-30% is quite normal in a second round. Does you study touch directly the gender effects? If there no special gender issues, you can consider your study to be valid(taking into account point 1).
