I would like to study antigen-specific T cell responses to an alphavirus (+ve sense RNA virus that replicates in the cytoplasm). Does anyone know whether it is best to engineer a virus that expresses the whole OVA protein, or can one just use the two epitopes (OVA323-339: ISQAVHAAHAEINEAGR and OVA257-264: SIINFEKL). If you can just make the two peptides, do they require a bridging sequence? We plan to use the virus in c57bl/6 mice that have been given OT-I or OT-II cells.
