We intend to conduct a project related to the effects of different oil sources and nitrate on ruminal some rumen parameters including methane production. With this aim, we will prepare a dairy cattle ration (2nd lactation). We will take a sample (0.2 g) and then insert in gas tight syringes which are used in in vitro gas production technique. And, we will determine the methane concentration in these gas tight syringes after 24 hours. But, there is a big problem. How we can obtain rumen content of a 2nd lactation cow??? We do not have a cannulated cow. Can I use rumen contents obtained from a different tytpe ruminant? Can you share your knowledge and experiences realted to this subject?

I think thats is better to canulate the cows. To use a fettening cow problabily will change the dry matter intake, and consequentilly, the methane production.

As a source of rumen inoculum, fattening cows can also be equally used as that of dairy cows provided uniformity of sampling and minimising variation between runs should be ensured that would yield consistent results in IVGPT.

...if diet of the both animals are same then we can use both. As a random sampling this can be used as a substitute of each other.

If you only need fluid for going into your gas chamber, you can get rumen fluid with a non-fistulated cow via a large needle (rumenocentesis) to seed your chamber.
Alternatively, a fistulated dairy cow might be available at a nearby institution if yours does not have one. It may be worth looking around for. I do agree that if nothing else, a fattening cow on a similar diet would be an alternative (but it may be difficult to find such an animal, as lactation diets are not typically used in this way).
In any cow, you can select the contents of the rumen with a stomach-tube and pump.
For an animal, this is a low-traumatic manipulation. If this is not done often. It is important how much fluid you need.
To semple a small volume of rumen fluid (5-8 ml), we use a special device (patented). Device is easy to input per os and without using a pump.
I have taken ruminal fluid indiscriminately to digest food in both dairy cows and fattening cattle and I have obtained similar results

Direct answer: no, because of the diet you do not. However in some particular situations, it depends on the diet you feed your dairy cows.
If your dairy cows are eating exactly the same diet than your fattening cows, you can use rumen contents of your fattening cow to estimate (not to determining) the rumen parameters of your dairy cow.
I agree with Mr. Partha Sarathi Swain , the key is to use the same feed for the cannulated animal and have a prudent period of adaptation to the diet
I feel you can use in the above said condition. Irrespective of the animal you need the innoculum and rumen fluid to see the effect. But if you want to study the effect in beef animals then you sld focus on beef rumen liquor only. As your study title becomes specific. If this is generalised you can sue both.

On my opinion, you have not need to fistulate cows; you have not need to use rumen fluid from fattening cows; you need to have rumen fluid per os by probe! But, let a veterinarian to do this - to put the probe in the right place - through the esophagus, and avoid the trachea! Have a good success.

I think you can use the bulls to see rumen envirioment changes due the treatment you use, remember that many of the clasic experiment on rumen enviroment were done in sheep and them carefully extrapoled to other ruminats; latter the selected treatment should be tested in cow if milk production features is into the the scope of your research. For gas in vitro test you also can use voided feaces from bulls. best regards,

Saludos, segun mi experiencia si se puede y da muy buenos resultados. La técnica in vitro propuesta por el Dr. Menken "tecnica de producción de gas" utiliza contenido ruminal de animales canulados o animales recien sacrificados en mataderos, ese contenido ruminal gaseado en CO2 y en preciencia de nutrientes (minerales y sales) se coloca a 39oC a incubar y puesdes estimar degradabilidad de la materia seca, pH, materia orgánica, AGV, CH4, CO2, CO, biomasa microbial, y parámetros de la cinetica ruminal; a, a+b y c...lagtime....revisa mis publicaciones.
