I'm planning to make f/2-silica medium for culturing my nannochloropsis. because of the time limit, i cannot use membrane filter sterilization, so that i come up to autoclave it. I want to know if there is any change in my media about the vitamin, the mineral and pH of the f/2-silica medium.

You can autoclave all the F/2 solutions (nitrate, phosphate, metals and silicates) but you can not autoclave vitamins. With the heat, vitamins will denature. If you are time-limited, filter just the vitamin solution and then added it to the final solution (composed by the other autoclaved solutions) in a flow chamber or in a steril environment.

sure you can, no problem. However, why a silicate-enriched medium for Nannochoropsis? Last I saw it does not need this enrichment., or at least, the amount present in natural seawater or leaked from glass flasks will be plenty, if traces of Silicon are needed in some physiological process.

I misunderstood, so what your question meant was f/2 less dilicon. Most manuals will tell you that vitamins should be filter-sterilized, but if you really wish, you can try comparing medium added filter-sterilized vitamins to complete medium autoclave-sterilized. In this case, you will have your experimental answer. otherwise, you may believe somebody who played with algae cultures for about 40 years, and autoclave you complete medium. As far as I can remember, and in spite of my >70 years nobody has added Alzheimer to my several age-related problems, I have never found a significant difference in growth and yield using autoclave-sterilized medium (that includes Nannochloropsis cultures)
woow IC. yeah i have try for about 3 day. btw the autoclaved medium used to have some white precipitate, while the unautoclaved didn't. But, indeed sir, the relative ratio of OD between the unautoclaved and autoclaved is about the same. thanks so much sir for your answer. this will be my basis for the next step in my research. thanks so much.
oops, sorry, I missed your answer. Since there are no silicates (which will not precipitate, although they might polymerize and give a slightly opalescent appearance to the medium) the only reason for seeing a precipitate after autoclaving would be the presence of phosphates, which might coprecipitate with metals.
just make sure that they will redissolve if you leave the medium standing a few hours. If they don't, check PO4 concentrations. Filtering off the precipitate and having it analyzed would also be a good idea. Loss is probably unimportant since you apparently have normal growth, but knowing as much as you can about the medium is the only good way to go.
