Biofilm is a fragile structure and can be easily disintegrated, so is there any method to make biofilm slides to study its architecture and morphology?
Thanks Shelley and Gabrielle for the suggestions. I would try out the PFA fixation method. But can you guys also elaborate how you generally grow biofilms? I have plans to grow biofilm on a coverslip whether is it a good method? Won't the biofilm disintegrate during the fixation and washing steps?
@Renu: I would recommend you to dig in "old" (1998-2005) articles of Paula Watnick
. They used a dye to quantify biofilm of a Pseudomonas biofilm. I used their approach long time ago but I clear remember it worked pretty well. Good luck. IB
