I do not see why you cannot extract your analyte from scrapings of the correct spot on your TLC plate. Use, let us say, chloroform or ether to wash the scrapings.  Put resulting washings in small droplets onto a glass slide while evaporating the solvent with a stream of air.   

Thanks for this suggestion..But they need minimum of 1mg samples. By scraping TLC spot getting only ng quantities.

This is why we began work with column chromatography when needing milligrams, way back in 1970.  Use the conditions to determine where your spot has hopefully travelled, and transfer the knowledge of polarity of solvent and column.  Then you check your collected column fractions with the TLC spot analysis to find the active fraction.
