Now I work and try to optimize the vehicle scheduling and routing problem using arena Simulation but need to know and learn other softwares and approaches.
We are using MATSim + DVRP extension to simulate and optimize on-line electric taxi dispatching. It is also used for simualting DRT (e.g. Univ. of Melbourne). You may find more on this here: http://www.vsp.tu-berlin.de/publications/vspwp/

This may help you. There is an interesting article that you can read (http://www.massey.ac.nz/~kahawick/cstn/095/cstn-095.pdf) with six software compared. Also, by experience the not free software every year come up with new and interesting features.
I agree with Isidro Ramos Torres -- I teach a university master's-level course in simulation using SImio, including for transportation; it has proved excellent, in power, ease of learning and use, and in quality and promptness of support.
I concur with Simio. It is very nice and powerful (it may be a bit overwhelming in the beginning, but once you get it, you will like it). Simul8 also could be interesting (very different than others)
