I am doing the DPPH assay and getting negative spectral value all the time. Can anybody suggest me another method or any explanation for the reason.

The ability to scavenge free radicals was estimated using DPPH radical as described by Ghatak et al, 2011. A 0.3mM solution of DPPH in methanol was prepared and 0.5mL of this solution was mixed with 100μL. The mixture was kept in dark at 37˚C for 30 min. The absorbance was measured spectrophotometrically at 517nm. The ability to scavenge DPPH radical was calculated using following formula,
Formula is: %inhibition = Abs of control - Abs 0f sample/Abs of control *100
