As the different stocks are prepared and mixed to prepare the final media, the trace metal solutions are not autoclaved rather filter sterilized. But i have autoclaved it and may be the sodium molybdate component is not behaving properly! plz suggest the exact steps anybody who has workbench knowledge in this.
When u prepare the stock solution, u must be careful with the micro nutrients. U add 1 ml of each micronutrient into 1.0 L dH2O. From there, u take 1mL into your final medium and of course macro nutrients and vitamins. Did I answer your question?
in fact, you may autoclave even vitamins (especially if you acidify them before sterilization). what do you mean with the sodium molybdate not behaving? if ypu get a precipitate (you shouldn't), add few drops of HCl and wait. it will redssolve
Thank you Domenico Voltolina sir! Actually i read somewhere that micronutrients and vitamins should not be autoclaved rather filter sterilized. But i hv autoclaved the micronutrients(trace metals solution). yeah sod. molybdate was precipitating , hence i autoclaved it separately and added to culture medium. i was worried for my results which were concurrent , but now its deviating. So to troubleshoot i am checking each possible aspect of my work.
correct. you should do that by all means, but if you think back, why was it working before and now it is deviating? to me this sounds as if something changed, but not in the preparation of the medium. So if the source of your chemicals is the same and the technique for medium preparation, sterilization etc is the same (check that: did you change brand', remember that, if you are dealing with diatoms, silicates may polymerize and become only very slowly available to your algae..etc), then to me you should start checking your original strain.
