i want to conduct an experiment on enzyme kinetic study on phosphate solubilizing bacteria on acid phosphatase enzyme. which plot shall i use and any papers or suggesstion for this?
The classical Lineweaver-Burk plot is good for the presentation of data, but unsuitable for the determination of Vmax and Km, as the taking of reciprocals changes the weight of the data points. Better suited is non-linear curve fitting to the original data, either by Runge-Kutta or (better) by Nelder-Mead. I have explained all this in doi:10.1007/978-3-319-19920-7_5.
The actual measurement is straight forward, either photometrically with para-nitrophenyl phosphate (pNPP) or (if the amount of enzyme is limiting) fluorimetrically with 4-methylumbelliferyl phosphate.
