why we need to use cumulative yield while plotting the graph of yield vs time instead of using non-cumulative? since for thermal labile compounds, the degradation (reducing of yield) observe for non-cumulative data vs time. this will describe the mechanisms of extraction rather than cumulative yield for thermal labile compounds. sorry for obvious misunderstanding on the concept of extraction compounds from plant.
Yes you can find a cumulative data on percent yield; ration of active principles to total components, heterocylic or simple compound ration, degree of isolation of organic compound and inorganic compound, solvent : product ration.
Good question, but technically difficult to address. When you monitor "extraction efficiency" you measure by certain metabolite group (say phenolics, sugars, alkaloids, terpenoids, steroids, amino acids, fat etc.), but when you wish to measure "what is degrading" then you need to monitor each of the groups separately and plot. But, stability studies (whether solvents causing compounds to degrade, break, modify, oxidise etc.) would be different from extraction efficiency (solvents promoting extraction of some group of metabolites) studies. Both have to be independent studies. RSM v(response surface methodology), ANN etc. models have been developed to monitor these parameters as well. best wishes.
thank you for your answer sir. please correct me if i am wrong. since the cumulative yield cannot determine the degradation of compounds, is that any possibility i determine the degradation by plotting non-cumulative yield vs time? but the difficulty that i am facing that the yield is inconsistent because of the complex structure of plant itself
Right. Then this is extraction problem. So extraction needs to be optimized with different solvents, time, acidification, temeprature, ratio of solvent: matrix etc. But then with time the yield decrease would not be degradation per se, but only reduced mount of components from matrix over time. To monitor degradation extraction should be continued with sampling by aliquoting at different time points, without removing the bulk solvent/ matrix (your plant materials), with concomitant monitoring of specific groups- say phenolics by Folin Ciocalcteau regaent, or sugar by phenol-sulfuric acid method and so on, or say 650 nm or so for chlorophyll degradation, 220 nm for peptides, 280 nm for proteins etc. You have to target a group of compounds by spectrophotometry to see which ones are degrading over time or conditions. thanks, Biswa
dear biswa, thank you for your answer. so u mean that should i use non-cumulative yield to investigate the degradation. example, i set up extraction time for 1, 3, 5..min then each time studied, i should use different sample and solvent namely at time 1min= sample1, time 3 min=sample 2 etc. then i can study degradation right?
