I have a groups of mutants (Mis) and another mutant (Mj), all derived from the same wild type. By competition experiments, I measured the fitness of Mis against Mj and that of Mj against wild type. I then then standardized the fitness of Mis against the wild type by multiplying their fitness with that of Mj's relative the wild type. This has been commonly done. Now I want to calculate the selection coefficient of the Mis against the wild type.
Anyone know a good equation for this?
On the one hand, the difference between 1 and the ratio you've inferred between Mis and Wt gives you a rough estimate, but to do a more thorough job allowing for the effects of error on your measurement and random genetic drift between the two mutant lines, I'd check out the method used by this great paper by Gallet et al 2012 in Genetics. http://www.genetics.org/content/190/1/175.full.pdf
