Dear fellow researchers,
In my experiment I would like to measure the influence of an electromagnetic field on the calcium flux, but want to minimize the other influences as much as possible.
After searching on Google and PubMed and not retrieving good results I'm searching for literature/keywords on how intracellular calcium signaling in (jurkat) cells is dependent on experimental setup. I'm thinking on temperature (RT under microscope instead of 37°C), starvation (change from medium to PBS), attachment surface.
Therefore I would like to know more about these influences on the cells. I would appreciate literature recommendations for research and review papers, as well as keywords on what I have to look up in PubMed or similiar.

My group dabbled with this type of work some time ago (Article: Exposure to GSM RF fields does not affect calcium homeostasis in human endothelial cells, rat pheocromocytoma cells or rat hippocampal neurons.
PLoS ONE 01/2010; 5(7):e11828). Some key issues are making sure that there is sufficient signal-to-noise, fidelity and reproducibility in your recordings that small changes in calcium signalling are evident. Some of the literature on effects of EMF on cells (including Jurkat cells) is simply too crude to be reliable in my opinion.
You are quite right that parameters such as temperature, presence of growth factors, substrate etc will alter how cells behave. What you have to do is define a minimal, controllable situation in which the cells are healthy and primed to respond in a robust and reproducible manner. You need to compare responses of control and test cells in the same incubator/chamber with your coils/magnet on and off, and the positions reversed. Double-blind experiments too, if possible.

I have some of these parameters in mind and some more, its defenitely necessary to control as much as possible in this area of research. We're hoping do add some really controlled experiments to the present pool of Jurkat experiments. Actually we're planning on a controlled incubator/chamber where all places should be equal.
